There was no relationship between the degree of activation of pyruvate, Pi dikinase, and adenylate energy charge, or ATP/ADP ratio or ADP level under various light intensities. Only a weak relationship was found between the degree of activation of NADP-malate dehydrogenase and the NADPH/NADP ratio or NADP(H) level with increasing light intensity. Abstract. Maize opaque2 (o2) mutations are beneficial for endosperm nutritional quality but cause negative pleiotropic effects for reasons that are not fully targets of the bZIP transcriptional regulator encoded by o2 include pdk1 and pdk2 that specify pyruvate phosphate dikinase (PPDK). This enzyme reversibly converts AMP, pyrophosphate, and phosphoenolpyruvate to ATP.
Pyruvate, Pi dikinase and NADP‐malate dehydrogenase (EC ) were the only enzymes examined which were light activated and increased in activity with increasing light intensity. In the low light grown plants the activity of pyruvate, Pi dikinase closely paralleled the photosynthetic rate measured under different light levels. And so this is kind of this second major use of Acetyl-CoA in the body. And so keep these kind of two major pathways for Acetyl-CoA in mind as we talk about how this step is regulated. Now I should say at this point that the major form of regulation, in this case, is allosteric regulation of the pyruvate dehydrogenase enzyme.
To investigate the feasibility of this, we generated a quadruple line that simultaneously accumulates four of the core C4 photosynthetic enzymes from the NADP-malic enzyme subtype, phosphoenolpyruvate carboxylase (ZmPEPC), NADP-malate dehydrogenase (ZmNADP-MDH), NADP-malic enzyme (ZmNADP-ME), and pyruvate phosphate dikinase (ZmPPDK). The activity and extent of light activation of three photosynthetic enzymes, pyruvate,Pi dikinase, NADP-malate dehydrogenase (NADP-MDH), and fructose 1,6-bisphosphatase (FBPase), were examined in maize (Zea mays var Royal Crest) leaves relative to the rate of photosynthesis during induction and under varying light was a strong light activation of NADP-MDH and Cited by: Pyruvate,Pi dikinase (PPDK, EC ) and NADP-malate dehydrogenase (MDH, EC ) were activated in the light and inactivated following a dark treatment in mesophyll protoplasts of maize.
DCMU (up to 33 micromolar), an inhibitor of noncyclic electron transport, inhibited activation of MDH much more strongly than it did by: There was a strong light activation of NADP-MDH and pyruvate,Pi dikinase, and light also activated FBPase 2- to 4-fold. During the induction period for whole leaf photosynthesis at 30°C under high light, the time required to reach half-maximum activation Cited by: Pyruvate,Pi Dikinase and NADP-Malate Dehydrogenase in C4 Photosynthesis: Properties and Mechanism of Light/Dark RegulationCited by: Regulation of the Galactolipid Synthesis in Spinach Chloroplasts.

Adenine Nucleotide Levels, the Redox State of the NADP System, and Assimilatory Force in Nonaqueously Purified Mesophyll Chloroplasts from Maize Leaves under Different Light Intensities | Plant Physiology. [Extract] C4 Cycle and its Significance in Photosynthesis This review considers two key enzymes of the C4 pathway of photosynthesis, pyruvate,Pi dikinase (PPDK) and NADP-malate dehydrogenase (NADPMDH).
I Besides their critical roles in C4 photosynthesis, these enzymes also share in common the fact that their activity is rapidly modulated by changes in light intensity. 1. Introduction. Historically speaking, plant pyruvate, orthophosphate dikinase (PPDK; E.C.
) was initially discovered in C 4 leaves, where it is an abundant mesophyll-chloroplast enzyme involved in C 4 photosynthesis.A major emphasis was placed on gaining an understanding of its now well-elucidated regulation via site-specific reversible phosphorylation of a target Thr residue.
Following preillumination in the presence of pyruvate, addition of 3-phosphoglycerate, oxaloacetate, or nitrite substantially diminished the activity of NADP-malate dehydrogenase.

NADP-malate dehydrogenase extracted from darkened leaves of the C3 plants pea, barley, wheat and spinach was activated by reduced glutathione, a monothiol, as well as by dithiothreitol (DTT).
However, in the C4 plants maize and Flaveria trinervia, only dithiothreitol could effectively activate the enzyme. There was no activation of the maize enzyme and little or no activation of the F.
Pyruvate, phosphate dikinase, or PPDK (EC ) is an enzyme in the family of transferases that catalyzes the chemical reaction.
ATP + pyruvate + phosphate ⇌ AMP + phosphoenolpyruvate + diphosphate. This enzyme has been studied primarily in plants, but it has been studied in some bacteria as well. It is a key enzyme in gluconeogenesis and photosynthesis that is responsible for reversing.

As pyruvate Pi-dikinase activities are only just high enough in NADPME-type species to support photosynthetic flux (Furbank et al., ), it is likely that this value could reflect the relative flux through the two decarboxylation enzymes.
It is currently unknown whether relative flux through these pathways is plastic or regulated by. Edwards GE, Nakamoto H, Burnell JN, Hatch MD.

Pyruvate,Pi dikinase exhibits complex regulation mediated by adenine nucleotides, Pi, and a high mol wt protein factor, but reductive activation by thiol/disulfide interconversion is not considered to be critical in the regulation of this enzyme (34).
